Results: The distribution of (18F)-PSS232 closely matched that of the well-characterized tracer (11C)-ABP688. After 45 minutes, tracer uptake was highest in anterior cingulate, basal ganglia and temporal cortex. Uptake was lowest in white matter and cerebellum. Metabolism of the tracer was well-fitted by a single exponential model with approximately 70% intact tracer after 90 min. Tracer kinetics were best described by a two tissue-compartment model. The extraction fraction was initially estimated at 50%. Conclusions: Brain uptake of (18F)-PSS232 matched the expected distribution of mGluR5 and kinetic modeling indicated that distribution volumes can be reliably estimated. A bolus/infusion protocol will be investigated and dosimetry measured.
